My advice would be leave it overnight as you will have to remove the spacers to actually grout and depending on how tight you have wedged the tiles to the spacers you may end up pulling off the tiles.
Mortar must set at least 24 to 48 hours before grouting.
Generally 24 hours will do for this and some setup more quickly.
Follow the package instructions for the tile mastic or thinset used for adhering the tiles to the wall.
